Overview

Pantothenate (vitamin B5) is the key precursor for the biosynthesis of CoA, an important cofactor involved in a variety of metabolic reactions including the citrate cycle and fatty acid biosynthesis. Pantothenate synthesis happens in plants, fungi and some bacteria, whereas animals salvage pantothenate from diet. In all living organisms, CoA is synthesized via a five-step process, which requires pantothenate and ATP as well as cysteine. In addition, CoA can be synthesized via alternate routes when intracellular CoA level are reduced and the in vivo pathway is impaired. The aberrant pantothenate and CoA biosynthesis therefore leads to human diseases like neurodegenerative diseases and cancers.
